As the sample is applied to the column, only the analyte and compounds close to it are retained by the column, while other components of the sample pass through unretained. After the analyte has bound to the column and other materials have been washed away, a wash buffer is applied and the analyte is eluted. After this wash step, the application buffer is reapplied to the column to allow for regeneration of the antibodies before another sample is applied.
